Part is designated as having issues because it appears to be lethal in cells containing the Tn10 transposon. The hypothesis is that the ptet promoter located on the high copy number plasmid "pulls" TetR created by the chromosomally encoded Tn10 transposon, causing massive derepression of TetA efflux pump, which is lethal in high levels.
